Immunocompromised mammalian hosts are subcutaneously implanted with a combination of human fetal bone and spleen. The chimeric animals can produce human B-cells, myeloid cells and T-cells for up to 9 months in vivo when supplied with an allogeneic human fetal thymic fragment at the same site. Grafts contain cell populations expressing CD4 and CD8, CD19 or CD33, CD14 and CD15, all of which also express the HLA type of the fetal bone/spleen. T-cells derived from progenitors in the fetal bone/spleen contain both mature single positive CD4+CD8-, CD8+CD4- as well as a high percentage of immature double positive CD4+CD8+ populations.
On place des implants sous-cutanés constitués d'un combinaison d'os et de rate de foetus humain chez des mammifères hôtes présentant un déficit immunitaire. Les animaux chimères peuvent produire des lymphocytes B, des cellules myéloïdes et des lymphocytes T humains pendant jusqu'à 9 mois in vivo lorsqu'ils reçoivent en même temps un fragment de thymus de foetus humain allogène, au même site. Les greffes contiennent des populations CD4 et CD8, CD19 ou CD33, CD14 et CD15, qui expriment toutes le type HLA des os/rate de foetus. Les lymphocytes T obtenus d'ascendants d'os/rate de foetus contiennent simultanément des populations CD4+CD8- et CD8+CD4- mûres et simplement positives, ainsi qu'un pourcentage élevé de populations CD4+CD8+ immatures et doublement positives.
