This invention relates to a process by which organic solvent-containing solutions are used in lieu of pure water for the preparation of cartilage extracts and fractions thereof. Different organic solvents have been tested for the preparation of extracts containing biologically active components possessing at least an activity against a matrix metalloprotease (anti-MMP). This invention also relates to a process by which a total liquid extract of shark cartilage composed of molecules having molecular weights less than about 500kDa (0-500 fraction) is purified into two well separated fractions composed of molecules having molecular weights less than about 1 kDa (0-1 fraction) and between about 1 to 500 kDa (1-500 fraction). In order to prevent the formation of aggregates, to improve the dissolution and the stability of active components, sucrose or other stabilizing agents can be added. The 0-500 fraction, the 0-1 fraction and the 1-500 fraction as well as their equivalents containing 1 % w/v sucrose possess anti-MMP and anti-tumor activities. The invention also provides a component which has at least anti-MMP and anti-tumor activities, and which has a mass of 244 amu (atomic mass unit). The use of the above extracts, derived fractions or components for inhibiting MMP enzyme, neovascularization and/or metastase formation is also described.

Cette invention a trait à un procédé dans le cadre duquel on utilise des solutions contenant un solvant organique à la place d'eau pure pour préparer des extraits de cartilage et leurs fractions. Différents solvants organiques ont été essayés aux fins de la préparation d'extraits contenant des composants biologiquement actifs témoignant d'au moins une action contre une métalloprotéase matricielle (anti-MMP). Cette invention concerne également un procédé dans le cadre duquel un extrait total liquide de cartilage de requin, composé de molécules dont les poids moléculaires sont inférieurs à environ 500 kDa (fraction 0-500), est purifié en deux fractions bien séparées composées de molécules dont les poids moléculaires sont inférieurs à environ 1 kDa (fraction 0-1) et de molécules dont les poids moléculaires ont une valeur comprise entre 1 et 500 kDa (fraction 1-500). Il est possible, afin d'éviter la formation d'agrégats, d'améliorer la dissolution et la stabilité des composants actifs, d'ajouter du saccharose ou d'autres agents de stabilisation. Les fractions, 0-500, 0-1, et 1-500 ainsi que leurs équivalents contenant 1 % p/v de sucrose ont une action anti-MMP et anti-tumorale. Cette invention porte, en outre, sur un composant doté d'au moins une action anti-MMP et anti-tumorale, ce composant ayant une masse de 244 u.m.a. (unités de masse atomique). L'invention concerne également l'utilisation des extraits susmentionnés, de fractions dérivées ou de composants afin d'inhiber l'action de l'enzyme MMP, la néovascularisation et la formation de métastases.
