The invention provides nucleic acids, peptides, and antibodies for use in applications including diagnosis and therapy. The peptides target lung cancer and were identified by phage display. Targeting phage PC5-2 and synthetic peptide SP5-2 were both able to recognize human pulmonary tumor specimens from lung cancer patients. In SCID mice bearing NSCLC xenografts, the targeting phage was able to target tumor masses specifically. When the peptide was coupled to liposomes containing the anti-cancer drugs vinorelbine or doxorubicin, the efficacy of these drugs against human lung cancer xenografts was improved, the survival rate increased, and the drug toxicity was reduced.
L'invention concerne des acides nucléiques, des peptides et des anticorps pour une utilisation dans des applications comprenant un diagnostic et un traitement. Les peptides ciblent le cancer du poumon et ont été identifiés par affichage de bactériophages. Le bactériophage de ciblage PC5-2 et le peptide de synthèse SP5-2 ont été tous deux capables de reconnaître des spécimens de tumeur pulmonaire humaine chez des patients atteints du cancer du poumon. Chez des souris SCID porteuses de xénogreffes, le bactériophage de ciblage a été capable de cibler spécifiquement des masses tumorales. Lorsque le peptide était couplé à des liposomes contenant les médicaments anticancéreux vinorelbine ou doxorubicine, l'efficacité de ces médicaments face aux xénogreffes du cancer du poumon humains était améliorée, le taux de survie accru et la toxicité médicamenteuse réduite.
