Vitamin D-binding protein (Gc protein) and its small domain (approximately 1/5 of the Gc peptide also known as domain III) were cloned via a baculovirus vector. The cloned Gc protein and the cloned domain (Cd) peptide were treated with immobilized .beta.-galactosidase and sialidase to yield macrophage activating factors, GcMAFc and CdMAF, respectively. These cloned macrophage activating factors and GcMAF may be used for therapy of cancer, HIV-infection and osteoporosis, and may also be used as an adjuvant for immunization and vaccination. Methods and an antibody-sandwich ELISA kit were developed to detect serum or plasma .alpha.-N-acetylgalactosaminidase in cancer and HIV- infected patients.
On a cloné à l'aide d'un vecteur de baculovirus la protéine de liaison de vitamine D (protéine Gc) et son petit domaine (égal au cinquième environ du peptide Gc, également appelé domaine III). On a traité la protéine Gc clonée et le peptide du domaine cloné (Cd) à l'aide de .beta.-galactosidase et de sialidase immobilisées, afin d'obtenir des facteurs d'activation de macrophages, respectivement le GcMAFc et le CdMAF. Ces facteurs clonés d'activation de macrophages et le GcMAF sont utilisables dans le traitement du cancer, de l'infection par le VIH et de l'ostéoporose, ainsi que comme adjuvant à des fins d'immunisation et de vaccination. On a élaboré des procédés et une trousse ELISA à sandwich anticorpal dans le but de détecter la .alpha.-N-acétylgalactosaminidase sérique ou plasmatique chez les malades atteints du cancer et chez les personnes infectées par le VIH.
