A method is described for the preparation, characterization and utilization of temporally ordered panels of cDNA libraries from human microclones. Each microclone undergoing morphogenetic differentiation in vitro differs in discrete gene expression. The developmental stage of the microclone is determined and the gene expression patterns of the microclones temporally ordered and arranged into expression profiles and temporal spectra. Direct comparison of the cDNAs produced from different neural microclones allows identification of novel RNA transcripts.
L'invention concerne un procédé de préparation, de caractérisation et d'utilisation de groupes classés selon un principe temporel de bibliothèques d'ADNc provenant de microclones humains. Chaque microclone, qui subit une différentiation morphogénétique in vitro, diffère dans l'expression des gènes discrets. On détermine le stade de développement du microclone, et les motifs d'expression sont classés selon un principe temporel puis regroupés en des profils d'expression et des spectres temporaux. Une comparaison directe des ADNc produits à partir de différents microclones neuraux permet d'identifier de nouveaux transcrits d'ARN.
