A substantially enriched mammalian hematopoietic cell population is provided, which is characterized as a progenitor cell committed to the megakaryocyte lineage. Methods are provided for the isolation and culture of these cells. The cell enrichment methods employ reagents that specifically recognize CD9 and CD41, in conjunction with other markers expressed on lineage committed progenitor cells. These cells give rise exclusively to megakaryocytes and platelets, as evidenced by their growth and differentiation in vitro and in vivo.
L'invention concerne une population de cellules hématopoïétiques mammifères sensiblement enrichies caractérisée comme étant une cellule souche participant à la lignée des mégakaryocytes. Des procédés sont prévus pour l'isolement et la mise en culture de ces cellules. Les procédés d'enrichissement cellulaire utilisent des réactifs reconnaissant spécifiquement CD9 et CD41, conjointement avec d'autres marqueurs exprimés dans des cellules souches participant à la lignée. Ces cellules donnent naissance exclusivement à des mégakaryocytes et des plaquettes mis en évidence par leur croissance et leur différenciation in vitro et in vivo.
