A process for the industrial manufacture in an aqueous medium of a slow- release excipient consisting mainly of cross-linked amylose having controlled release properties, for use in the preparation of tablets or pellets. A starch containing a high amount of amylose (high amylose starch) is first subjected to a gelatinization. The gelatinized high amylose starch is then cross-linked with 1 to 5 g of a cross-linking agent per 100 g of dry-based gelatinized high amylose starch in an alkali medium, thereby forming a reaction medium containing a reaction product consisting of a cross-linked high amylose starch slurry. The obtained reaction medium is neutralized, thereby forming by- products mainly consisting of salts, which are removed from the reaction medium. The recovered cross-linked high amylose starch slurry is then subjected to a thermal treatment at a temperature of at least 60 ~C and the thermally treated product is dried to obtain the requested slow release excipient. This process is economical and industrially advantageous since it is carried out in an aqueous medium.
Procédé de production industrielle dans un milieu aqueux d'un excipient à libération lente constitué principalement par de l'amylose réticulée possédant des propriétés de libération contrôlée, afin de l'utiliser dans la préparation de comprimés ou de granules. On soumet d'abord un amidon contenant une quantité élevée d'amylose (amidon à teneur élevée en amylose) à une gélification. On soumet ensuite cet amidon gélifié à une réticulation au moyen de 1 à 5 grammes d'agent de réticulation pour 100 grammes d'amidon gélifié à base sèche dans un milieu alcalin, ce qui permet d'obtenir un milieu réactionnel contenant un produit réactionnel consistant en une pâte liquide d'amidon réticulé à teneur élevée en amylose. On neutralise le milieu réactionnel obtenu, ce qui permet d'obtenir des produits secondaires constitués principalement par des sels, qu'on enlève du milieu réactionnel. On soumet ensuite la pâte liquide d'amidon réticulé récupérée à un traitement thermique à une température d'au moins 60 ·C et on sèche le produit traité thermiquement afin d'obtenir l'excipient voulu à libération lente. Ce procédé est économique et présente des avantages sur le plan industriel, étant donné qu'il est exécuté dans un milieu aqueux.
