A test method for providing data usable in predicting the onset possibility of acute graft-versus-host disease (GVHD); a kit for conducting this method; and a preparation and a molecule-targeting therapy for treating and preventing GVHD. The test method as described above comprises monitoring with the passage of time the blood DNAM-1 concentration of a patient having had hematopoietic stem cell transplantation from, for example, bone marrow and providing data for predicting the possibility of the onset of acute graft-versus-host disease, data for estimating the onset risk thereof or data for evaluating a therapeutic effect after the onset thereof based on changes in abnormal values exceeding the normal level. Moreover, a molecule-targeting therapy characterized by comprising targeting DNAM-1 molecule in the blood of a patient with the onset of GVHD or having a possibility of the onset thereof, administering anti-DNAM-1 antibody that is a neutralizing antibody therefor, and thus treating or preventing GVHD; and a preparation to be used therefor are disclosed.
L'invention concerne une méthode d'essai permettant de fournir des données utilisables dans la prédiction du risque de l'apparition de la maladie du greffon contre l'hôte (GVHD); un kit d'application de cette méthode; ainsi qu'une préparation et une thérapie de ciblage de molécule permettant de traiter et de prévenir la GVHD. La méthode d'essai de l'invention comprend la surveillance dans le temps de la concentration sanguine de la molécule DNAM-1 chez un patient ayant subi une transplantation de cellules souches hématopoïétiques, par exemple, issues de la moelle épinière; et l'obtention de données permettant la prédiction du risque de l'apparition de la maladie du greffon contre l'hôte, de données permettant d'estimer le risque de l'apparition de la maladie ou de données permettant d'évaluer un effet thérapeutique donné après l'apparition de la maladie, sur la base des modifications des valeurs anormales dépassant le taux normal. Par ailleurs, l'invention concerne une thérapie de ciblage de molécule caractérisée en ce qu'elle consiste à cibler la molécule DNAM-1 dans le sang d'un patient ayant développé la GVHD ou risquant de la développer, à lui administrer un anticorps anti-DNAM-1 pour la neutraliser, et par conséquent, traiter ou prévenir la GVHD. L'invention concerne en outre une préparation utilisée dans cette méthode.
