The subject invention concerns the novel use of formyl-CoA transferase enzyme together with oxalyl-CoA decarboxylase enzyme for the detection and measurement of oxalate in biological samples. The use of enzyme system according to the subject invention results in the conversion of oxalate into carbon dioxide and formate. Because the production of formate is directly correlated to the concentration of oxalate present in a sample, the determination of the resulting formate concentration provides an accurate, sensitive and rapid means for detecting even low levels of oxalate. The subject invention further concerns the cloning, sequencing and expression of the genes that encode the formyl-CoA transferase enzyme and the oxalyl-CoA decarboxylase enzyme of Oxalobacter formigenes. The subject invention also concerns a method for detecting the presence of Oxalobacter formigenes organisms in a sample, and the polynucleotide probes and primers used in the detection method.

L'invention concerne une nouvelle utilisation de l'enzyme formyl-CoA transférase conjointement avec l'enzyme oxalyl-CoA décarboxylase pour la détection et la mesure d'oxalate dans des échantillons biologiques. L'utilisation du système enzymatique selon l'invention permet de convertir l'oxalate en dioxyde de carbone et en formiate. La production de formiate étant directement liée à la concentration d'oxalate présente dans un échantillon, la détermination de la concentration de formiate résultante fournit un moyen sensible et rapide de détection de l'oxalate même en faible quantité. L'invention porte également sur le clonage, le séquençage et l'expression des gènes codant les enzymes formyl-CoA transférase et oxalyl-CoA décarboxylase d'Oxalobacter formigènes. L'invention se rapporte également à un procédé de détection de la présence d'organismes Oxalobacter formigènes dans un échantillon, ainsi qu'aux sondes et amorces polynucléotidiques utilisées à cet effet.
