This application discloses that inositolphosphoglycans (IPGs) can be obtained from basophils, eosinophils and mast cells and that allergen stimulation of these cells results in IPG release. It also shows that IPGs are second messengers for allergic stimulation as the addition of some types of purified IPGs to non-allergen stimulated cells resulted in histamine release or degranulation. Thus, IPG antagonists can be used for the treatment of conditions (especially allergy and asthma) mediated by the release of IPGs from mast cells, basophils or eosinophils. Preferred IPG antagonists include anti-IPG antibodies, inhibitors of the enzyme GPI-PLD and competitive antagonists.
La présente invention concerne la découverte de la possibilité d'obtenir des inositolphoglycanes (IPG) à partir de basophiles, éosinophiles et mastocytes et du fait que la stimulation allergène de ces cellules entraîne la libération d'IPG. La présente invention démontre également que les IPG sont des messagers secondaires d'une stimulation allergique puisque l'ajout de certains types d'IPG purifiés à des cellules à stimulation non allergène entraîne une libération d'histamine ou une dégranulation. Ainsi, on peut utiliser des antagonistes d'IPG pour traiter des troubles (en particulier une allergie ou de l'asthme) provoqués par la libération d'IPG de mastocytes, basophiles ou éosinophiles.
