Succinic amide derivatives of formula (I), wherein W is a -CO2H or -CONHOH group; R, R1 and R2 are each hydrogen or an organic residue, R3 is the residue of an alpha-aminoacid and R4 is an organic group, are inhibitors of matrix metalloproteinases (MMPs) and of the release of tumor necrosis factor-alpha (TNF) from cells, and are therefore useful in the prevention, control and treatment of diseases in which MMPs or TNF are involved. A process for their preparation and pharmaceutical compositions containing them are also described.

L'invention concerne des dérivés d'amide succinique présentant la formule (I). Dans la formule, W est un groupe -CO¿2?H ou -CONHOH; R, R¿1?, et R¿2? sont chacun de l'hydrogène ou un résidu organique, R¿3? est le résidu d'un acide alpha-aminé et R¿4? est un groupe organique. Ces dérivés sont des inhibiteurs de la métalloprotéinase matricielle et de la libération du facteur alpha de nécrose tumorale à partir des cellules. Par conséquent, ils sont très utiles pour prévenir, contrôler et traiter les maladies dans lesquelles les métalloprotéinases matricielles ou le facteur de nétrose tumorale jouent un rôle important. L'invention traite aussi de la préparation de ces dérivés et de compositions pharmaceutiques les contenant.
