The invention provides a use of a compound capable of at least in part influencing an activity of a neural reflex pathway for the preparation of a medicament for altering the motility of the gastrointestinal tract. Said medicament can be used for prophylaxis and/or treatment of hypomotility of the gastrointestinal tract. Preferably, generalized hypomotility occurring during postoperative ileus is prevented and/or treated by a medicament of the invention.In one aspect of the invention said activity of said neural reflex pathway is decreased by at least in part preventing stimulation of said pathway by an immunocyte, a macrophage and/or a mast cell. Said activity can also be decreased by at least in part preventing immunocyte recruitment. A compound capable of at least in part influencing an activity of a neural reflex pathway for instance comprises an anti-ICAM-1 antibody, an anti LFA-1 antibody, and/or ketotifen. A pharmaceutical composition for prophylactic and/or therapeutic treatment of an individual against hypomotility of the gastro-intestinal tract comprising a compound capable of at least in part decreasing a neural reflex pathway is also herewith provided.
L'invention se rapporte à l'utilisation d'un composé permettant d'influencer au moins partiellement l'activité d'une voie réflexe neuronale aux fins de la préparation d'un médicament permettant de modifier la motilité du tractus gastro-intestinal. Ledit médicament peut être utilisé pour la prophylaxie et/ou le traitement de l'hypomotilité du tractus gastro-intestinal. De préférence, un médicament de l'invention permet de prévenir et/ou de traiter une hypomotilité généralisée se produisant du fait d'un iléus postopératoire. Dans un mode de réalisation de la présente invention, on réduit l'activité de ladite voie réflexe neuronale en empêchant au moins en partie la stimulation de ladite voie par un immunocyte, un macrophage et/ou un mastocyte. Il est également possible de réduire cette activité en empêchant au moins en partie le recrutement des immunocytes. Un composé permettant d'influencer au moins en partie l'activité d'une voie réflexe neuronale comporte par exemple un anticorps anti-ICAM-1, un anticorps anti-LFA-1 et/ou un kétotifène. L'invention se rapporte également à une composition pharmaceutique permettant d'administrer à un sujet un traitement prophylactique et/ou thérapeutique contre l'hypomotilité du tractus gastro-intestinal et comportant un composé apte à réduire au moins en partie une voie réflexe neuronale.
