The invention provides means and methods for efficiently cloning nucleic acid sequences of interest in micro-organisms that are less amenable to conventional nucleic acid manipulations, as compared to, for instance, E.coli. The present invention enables high-throughput cloning (and, preferably, expressing) of a nucleic acid of interest in a first kind of micro-organism, while an initial (preferably high throughput) cloning of the nucleic acid of interest is done in a second kind of micro-organism.

L'invention concerne des moyens et des procédés pour cloner efficacement des séquences d'acides nucléiques d'intérêt dans des micro-organismes qui sont moins favorables aux manipulations d'acide nucléique classiques, par comparaison, par exemple, à E. coli. La présente invention permet le clonage à rendement élevé (et, de préférence, l'expression) d'un acide nucléique d'intérêt dans un premier type de micro-organisme, alors qu'un clonage initial (de préférence à rendement élevé) de l'acide nucléique d'intérêt est réalisé dans un deuxième type de micro-organisme.
