A method of determining the presence or onset of capillary leakage comprises introducing into the blood stream of the subject a first low dosage of a low molecular weight macromolecular material and determining the volume of distribution thereof, referred to herein as an apparent volume of distribution. The determined value of volume of distribution is influenced not only by extravasation through the capillary wall but also by binding of smaller molecules to pores or receptor binding sites in the wall which occurs much faster than extravasation and is an important indicator of the onset of capillary leakage. The process is repeated with a high dose of high molecular weight macromolecular material to determine a further volume of distribution referred to herein as the time circulating plasma volume. The two values are compared to determine the presence or onset of capillary leakage.
L'invention concerne un procédé permettant de mettre en évidence la présence ou l'apparition d'une fuite capillaire. Ledit procédé consiste à introduire dans le courant sanguin du sujet une première dose, faible, d'une matière macromoléculaire à faible poids moléculaire et à déterminer son volume de distribution, ici appelé volume de distribution apparent. La valeur du volume de distribution ainsi déterminée est fonction non seulement de l'extravasion à travers la paroi du capillaire mais aussi de la fixation de petites molécules sur les pores ou les sites de fixation de récepteurs situés sur la paroi, phénomène qui est beaucoup plus rapide que l'extravasion et qui constitue un indicateur important de l'apparition d'une fuite capillaire. On répète le processus avec une dose élevée de matière macromoléculaire à haut poids moléculaire afin de calculer un autre volume de distribution, ici appelé volume plasmatique circulant dans le temps. On compare les deux valeurs afin d'établir la présence ou l'apparition d'une fuite capillaire.
