This disclosure provides an improved system for deriving culturing human pluripotent stem cells. Traditionally, pluripotent stem cells are cultured on a layer of mouse embryonic fibroblast feeder cells to prevent them from differentiating. In the system described here, the role of feeder cells is replaced by defined components added to the culture environment that support rapid proliferation without differentiation. The medium contains an isotonic buffer, a blend of essential nutrients such as protein and lipids, and an effective growth factor or combination of factors that promote proliferation while inhibiting differentiation. Culturing human embryonic stem cells in fresh medium on an extracellular matrix according to this invention causes the cells to expand surprisingly rapidly, while retaining the ability to differentiate into cells representing all three embryonic germ layers. This new culture system allows for bulk proliferation of pPS cells for commercial production of important products for use in drug screening and human therapy.

L'invention concerne un système amélioré de culture de cellules souches pluripotentes humaines. Traditionnellement, on cultive les cellules souches pluripotentes sur une couche de cellules nourricières de fibroblastes embryonnaires de souris pour éviter qu'elles ne se différencient. Dans le système décrit, le rôle des cellules nourricières est remplacé par des composants définis ajoutés au milieu de culture qui permet une prolifération rapide sans différenciation. Le milieu contient un tampon isotonique, un mélange d'éléments nutritifs essentiels, tels que des protéines et des lipides, et un facteur de croissance efficace ou une combinaison de facteurs qui favorisent la prolifération tout en inhibant la différenciation. Selon cette invention, la culture de cellules souches embryonnaires humaines en milieu frais, sur une matrice extracellulaire, entraîne l'expansion étonnamment rapide des cellules tout en conservant leur aptitude à se différencier en cellules représentant les trois feuillets de germes embryonnaires. Ce nouveau système de culture permet d'obtenir une prolifération globale de cellules pPS à des fins de production commerciale de produits importants pour le criblage de médicaments et la thérapie humaine.
