An immunotherapeutic vaccine providing antigen presenting cells that have been pulsed with a disrupted cell preparation which includes enucleated cytosol and cell membranes of cancer cells infected with a recombinant vaccinia virus encoding at least one immunostimulating molecule. In a preferred embodiment, the vaccine includes autologous dendritic/monocytic cells (DC/M) that present a mixture of antigens (present in the enucleated cytosol and cell membranes) from melanoma cell lines that have been infected with a recombinant vaccinia virus encoding IL-2. In another of the preferred embodiments, the enucleated cytosol and cell membranes are from melanoma cells harvested from the patient to be treated. A method of making the vaccine and methods of using the vaccine to stimulate an anti-cancer immune response and to treat a patient with a cancer are also described.

L'invention concerne un antigène produisant un vaccin immunothérapeutique présentant des cellules pulsées par une préparation cellulaire interrompue comprenant des membranes cytosoliques et cellulaires énucléées de cellules cancéreuses infectées avec un virus vaccinal recombiné codant pour au moins une molécule immunostimulante. Dans un mode de réalisation préféré, le vaccin comprend des cellules dendritiques/monocytiques autologues (DC/M) qui présentent un mélange d'antigènes (présents dans les membranes cytosoliques et cellulaires énucléées) de lignée cellulaire de mélanome infectés avec un virus vaccinal recombiné codant pour IL-2. Dans un autre mode de réalisation préféré, les membranes cytosoliques et cellulaires énucléées proviennent de cellules de mélanome récoltées chez le patient à traiter. L'invention concerne également un procédé de fabrication de ce vaccin et des procédés d'utilisation du vaccin pour stimuler une réponse immunitaire anticancéreuse et pour traiter un patient atteint d'un cancer.
