The present invention relates to membrane-permeable peptides which are intracellular agonists and/or antagonists of chemotactic factor receptors are rendered hydrophobic through acylation and acetoxymethylation of their amine and acid functional groups. The modified peptides of the present invention are loaded into cells. The acetoxymethyl esters are cleaved by non-specific esterases rendering the peptides active in the intracellular compartments of the cells. The effects of the introduction of transformed specific peptides corresponding to intracellular regions common to the major chemokine receptors are illustrated. These peptides completely inhibited chemotactic factor and chemokine-induced calcium mobilization. Furthermore, leukocytes of mice intravenously injected with these peptides failed to migrate towards chemokines(IL-8).

La présente invention concerne des peptides à perméabilité membranaire qui sont des agonistes et/ou des antagonistes intracellulaires des récepteurs du facteur chémotactique. Ces peptides sont rendus hydrophobes par acylation et acétoxyméthylation de leurs groupes fonctionnels amine et acide. Les peptides modifiés selon l'invention sont chargés dans les cellules. Les esters d'acétoxyméthyle sont coupés par des estérases non spécifiques rendant les peptides actifs dans les compartiments intracellulaires des cellules. L'invention traite des effets de l'introduction des peptides spécifiques transformés correspondant aux zones intracellulaires communes aux principaux récepteurs de chémokine. Ces peptides ont totalement inhibé le facteur chémotactique et la mobilisation calcique induite par la chémokine. En outre, les leucocytes de souris dans lesquels on a injecté par voie intraveineuse ces peptides n'ont pas présenté de migration vers les chémokines (IL-8).
