The complete DNA sequences for adhE and pf1 genes of Lactococcus lactis, recombinant replicons comprising one or both of these genes or comprising mutants or variants hereof including mutants in which the genes are inactivated, and recombinant lactic acid bacteria comprising such a replicon are provided. The gene sequences and/or sequences regulating the expression of the genes can be modified to provide metabolically engineered lactic acid bacteria which have an enhanced or reduced production of one or more metabolites resulting from citrate and/or sugar fermentation. Such metabolically modified cells are useful as starter cultures in the manufacturing of food products and animal feed having improved flavour and/or shelf life, including dairy products, or they can be used directly in the manufacturing of a lactic acid bacterial metabolite.
L'invention concerne les séquences d'ADN complètes des gènes adhE et pfl de Lactococcus lactis, des réplicons recombinants contenant un de ces gènes ou les deux, ou leurs mutants ou variants, notamment des mutants dans lesquels les gènes sont inactivés, ainsi que des bactéries lactiques recombinantes contenant un réplicon de ce type. Les séquences géniques et/ou les séquences régulant l'expression des gènes peuvent être modifiées de façon à donner des bactéries lactiques à métabolisme modifié, qui produisent de manière augmentée ou diminuée un ou plusieurs métabolites résultant de la fermentation du citrate et/ou du sucre. Les cellules ainsi modifiées sont utiles comme cultures starters pour la fabrication de produits alimentaires et d'aliments pour animaux ayant un goût amélioré et/ou une durée de conservation accrue, notamment les produits laitiers, ou peuvent être utilisées directement pour la fabrication d'un métabolite des bactéries lactiques.
