A metabolically engineered micro-organism has an operative first metabolic pathway in which a first metabolite is transformed into a second metabolite in a reaction in which NAD is a cofactor for a first enzyme, suitably a phosphorylating dehydrogenase, said reaction step producing NADH. Said second metabolite is transformed into at least one further metabolite in a reaction catalysed by a second enzyme, suitably a kinase. The organism has an operative second metabolic pathway characterised by an enzyme activity in excess of a native level in respect of a third enzyme, suitably a non-phosphorylating dehydrogenase, e.g. GAPN, catalysing a non-reversible reaction in which NADP is a cofactor and NADPH is a product. Said first metabolite is transformed into a said further metabolite without the involvement of said second enzyme.

L'invention concerne un micro-organisme produit par ingénierie métabolique ayant une première voie métabolique opérationnelle, dans laquelle un premier métabolite est transformé en un second métabolite dans une réaction, au cours de laquelle NAD désigne un cofacteur pour une première enzyme, notamment une déshydrogénase de phosphorylation, ladite réaction produisant un NADH. Ledit second métabolite est transformé en au moins un autre métabolite dans une réaction catalysée par une seconde enzyme, de préférence une kinase. L'organisme présente une seconde voie métabolique opérationnelle caractérisée par une activité enzymatique excessive d'un niveau indigène par rapport à une troisième enzyme, notamment une déshydrogénase de non phosphorylation, p.ex. GAPN, catalysant une réaction irréversible, au cours de laquelle NADP désigne un cofacteur et NADPH désigne un produit. Ledit premier métabolite est transformé en un autre métabolite sans qu'une seconde enzyme ne soit impliquée.
