According to an embodiment of the present invention, a method for determining a conformation for a molecular structure is provided. The method includes a variety of steps such as computationally decomposing the molecular structure into fragments. A step of normalizing each of the fragments in order to form normalized fragments is also included. The step of determining at least one or possibly many conformers for each normalized fragment is included in the method. Finally, the step of combining at least a first conformer and a second conformer in order to derive the molecular structure is performed. Other conformers may also be included in the combination. Some embodiments will also include a step of searching for one or more conformers fragments in a library. If the fragment is found in the library, then its corresponding conformers are read from the library. Otherwise, the method includes an additional step of storing the fragment and conformer information it produces in the determining step into the library for subsequent analyses. Select embodiments according to the invention can produce low energy conformers by resolving strain and torsion within chemical bonds between atoms in the fragments.
Un mode de réalisation de la présente invention prévoit un procédé de détermination d'une conformation d'une structure moléculaire. Ce procédé comporte différentes étapes: une étape de décomposition informatique de la structure moléculaire en fragments; une étape de normalisation de chacun des fragments afin de former des fragments normalisés; une étape de détermination d'au moins un ou, si possible, de plusieurs conformères pour chaque fragment normalisé; et enfin, une étape de combinaison d'au moins un premier conformère et d'un second conformère afin de dériver la structure moléculaire. On peut ajouter à cette combinaison d'autres conformères. Certains modes de réalisation comportent également une étape consistant à rechercher un ou plusieurs fragments de conformères dans une banque. Si l'on trouve le fragment dans la banque, alors ses conformères correspondants sont extraits de la banque. Sinon, le procédé comporte une étape supplémentaire de stockage des informations relatives aux fragments et aux conformères qu'il produit à l'étape de détermination dans la banque en vue d'analyses ultérieures. Des modes de réalisation sélectionnés selon l'invention peuvent produire des conformères de faible énergie par résolution des souches et des torsions à l'intérieur des liaisons chimiques entre atomes dans les fragments.
