The invention relates to a method for the electrophoretic separation of particles, especially of macromolecules. The inventive method comprises the following steps: applying the particles to be separated on a substrate- supported membrane so that the particles are mobile across the surface of the substrate-supported membrane, supplying an electrical field in such a manner that electrical fields are formed across the surfaces across which the particles are mobile, temporarily modifying the electrical fields and/or using a substrate-supported membrane with a structured surface, the direction and strength of the electrical field being temporarily modified such and/or the substrate-supported membrane being structured such that a force is acting on the particles that leads to a movement that depends on the length of the particles. The invention further provides a substrate especially for supporting a membrane that consists of an optically transparent material and that is used in the inventive method. The invention is also characterized by a substrate-supported membrane for carrying out the inventive method with an inventive substrate according to any of the claims, and a fluid lipid membrane. The invention provides a micro-channel electrophoresis chamber with at least one channel the bottom of which comprises an inventive substrate and with an electrode system.
La présente invention concerne un procédé de séparation électrophorétique de particules, en particulier de macromolécules, comprenant les étapes suivantes: mise en place des particules à séparer dans une membrane supportée par un substrat, de sorte que les particules peuvent se déplacer le long de la surface de la membrane supportée par un substrat; apport d'un champ électrique de manière à faire se former des champs E le long de la surface le long de laquelle les particules peuvent se déplacer; et modification temporaire du champ électrique et/ou utilisation d'une membrane supportée par un substrat présentant une surface structurée. La direction et l'intensité du champ électrique sont modifiées temporairement de sorte que et/ou la membrane supportée par un substrat a une structure telle qu'une force agit sur les particules pour leur donner un mouvement dépendant de la longueur des particules. Cette invention concerne également: un substrat servant en particulier à supporter une membrane lors de la mise en oeuvre du procédé de l'invention, ledit substrat étant constitué de matériau optiquement transparent; une membrane supporter par un substrat, servant à la mise en oeuvre du procédé de l'invention, présentant un substrat, selon l'une des revendications, et une membrane lipidique fluide; une chambre d'électrophorèse à microcanaux comportant au moins un canal dont la surface du fond est constituée du substrat de l'invention, et un dispositif d'électrodes.
