The invention concerns the manufacture of electronic micro-sensors (chips) for analysing nucleotide and/or peptide sequences. The object is to fix at the surface of said chips a very large number of various probe sequences easily and economically. The method aims at making arrays of sequence probes on a support by anchoring and/or local chemical synthesis in the presence of volatile solvents. Said method is characterised in that it consists in using a support whereon is produced a grid of reaction wells made of light-cured (epoxy) resin; in placing the support in a reaction chamber which is saturated with vapours of the synthesis solvent (acetonitrile); in finally producing the anchorage and optionally the localised chemical synthesis in situ of the probe sequences in the wells using a localised supply of liquids/reagents in each well corresponding to a specific probe-sequence, and in supplying collectively the wells with common liquids/reagents and in ensuring that prior to each localised supply operation the liquid contained in each well is eliminated. The invention also concerns a device for implementing said method.
L'invention concerne la fabrication de microcapteurs ("puces") électroniques pour l'analyse de séquences nucléotidiques et/ou peptidiques. Le but est de fixer à la surface de ces "puces" un très grand nombre de séquences sondes différentes, de manière aisée et économique. Le procédé de fabrication de réseaux de séquences sondes sur un support par ancrage et/ou synthèse chimique localisé(s) en présence de solvants volatils. Selon ce procédé, on met en oeuvre un support sur lequel est réalisé un quadrillage de puits réactionnels en résine photoréticulée (époxy). On place ensuite le support dans une enceinte réactionnelle que l'on sature à l'aide de vapeurs du solvant de synthèse (acétonitrile). On réalise enfin l'ancrage et éventuellement la synthèse chimique localisé(s) in situ des séquences sondes dans les puits en mettant en oeuvre une alimentation localisée en liquides/réactifs dans chaque puits correspondant à une séquence-sonde donnée, en alimentant collectivement les puits en liquides/réactifs communs et en faisant en sorte que préalablement à chaque opération d'alimentation localisée le liquide contenu dans chaque puits soit éliminé. L'invention a également pour objet un dispositif pour la mise en oeuvre de ce procédé.
