It is intended to provide a method of releasing the transcriptional regulation caused by a repeated sequence in a gene, a kit therefor and so on to thereby establish a line capable of producing a protein in a large amount. The above- described method can be achieved by any one or more of the following methods: (a) in the amplification of a gene encoding a target protein, co-amplifying a polynucleotide of 10 kbp or more such as a .lambda.-phage DNA or an insulator sequence; (b) culturing cells having undergone gene amplification in media containing a drug with a gradual increase in concentration followed by screening; (c) elevating the promoter activity of inducing the expression of a gene encoding a target protein; (d) excising an amplified gene region from a chromosome with the use of Cre-LoxP System; (e) treating cells having undergone gene amplification with 5-aza-2~-deoxycytidine to thereby lower the methylation degree of DNA; and (f) selecting the mammalian cells having undergone gene amplification on double minute chromosomes.
L~invention concerne un procédé de libération de la régulation traductionnelle induite par une séquence répétée dans un gène, un kit pour celui-ci etc., afin de créer une lignée capable de produire une protéine en grande quantité. Le procédé décrit ci-dessous peut être obtenu par un ou plusieurs des procédés suivants : (a) dans l~amplification d~un gène codant pour une protéine cible, co-amplification d~un polynucléotide de 10 kbp ou plus tel qu~un ADN .lambda.-phage ou une séquence isolante (b) mise en culture de cellules ayant subi une amplification de gènes dans un milieu contenant un médicament avec augmentation progressive de la concentration suivie par un criblage (c) augmentation de l~activité de stimulation ou d~induction de l~expression d~un gène codant pour une protéine cible ; (d) extraction d~une région génétique amplifiée d~un chromosome au moyen du système Cre-LoxP ; (e) traitement des cellules ayant subi une amplification génétique par la 5-aza-2~-deoxycytidine afin de réduire le degré de méthylation de l~ADN ; et (f) sélection des cellules mammifères ayant subi une amplification génétique sur des chromosomes minuscules doubles.
