The object is to remove substances that can inhibit nucleic acid amplification from biological samples and nucleic acid extracts, and to allow convenient and accurate evaluation of the presence or absence of a target nucleic acid or the expression level of a target gene in a biological sample, by nucleic acid amplification means employing an enzyme. The invention provides a method for preparation of a sample for use in nucleic acid amplification, to be used for amplification of nucleic acid in a biological sample, which method comprises an extraction step in which a nucleic acid extraction reagent is added to the biological sample to obtain a nucleic acid extract, and a purification step in which the nucleic acid extract is contacted with zeolite to obtain a nucleic acid amplification sample suitable for nucleic acid amplification, to allow nucleic acid detection at high sensitivity.
L'objet de cette invention est d'éliminer toute substance qui inhibe l'amplification d'un acide nucléique contenu dans un échantillon biologique et une solution d'extraction d'acide nucléique, et d'évaluer avec précision et de manière simple la présence ou l'absence d'un acide nucléique cible ou le niveau d'expression d'un gène cible dans un échantillon biologique avec un moyen d'amplification enzymatique d'acide nucléique. Par conséquent, cette invention décrit un procédé pour préparer un échantillon en vue d'une amplification d'acide nucléique utilisable pour amplifier un acide nucléique contenu dans un échantillon biologique, qui comprend les étapes suivantes : une étape d'extraction consistant à ajouter un réactif d'extraction d'acide nucléique à l'échantillon biologique pour produire une solution d'extraction d'acide nucléique ; et une étape de purification consistant à mettre en contact la solution d'extraction d'acide nucléique avec une zéolite pour produire un échantillon en vue d'une amplification d'acide nucléique utilisable pour amplifier l'acide nucléique et qui permet la détection très sensible d'un acide nucléique.
