-0- METHOD AND KITS FOR DETECTING HUMAN LEUKOCYTE ANTIGEN DNA Abstract of the Disclosure A rapid and simple method for detecting HLA DNA comprises amplifying HLA DNA using polymerase chain reaction in which at least one primer is bio- tinylated, contacting denatured amplified DNA strands with an oligonucleotide probe, and detecting the insolubilized materials using a peroxidase-avidin conjugate. The probe comprises a polymeric particle and an oligonucleotide which is complementary to the amplified product, is 15 to 25 nucleotides in length, and is attached to the particle through a linking group having 16 to 30 ethylene glycol units. This method is used to advantage to type HLA, or to detect HLA DNA in a specimen. The materials needed to carry out the method can be provided in test kits. Desir- ably, the assay is carried out using a disposable test device having a microporous filtration membrane.
