ABSTRACT The chain DNA (A) comprising the plural deoxyribonucleotide and the primer of said chain DNA, preferably a primer comprising a base sequence of TCCAGCGGCGGG or AGGTCGCCGCCC, are prepared. The single stranded DNA (D) obtained by applying the thermal denaturation to this chain DNA is subjected to anneal with said primer. The complementary chain (F) is synthesized with the chain DNA annealed by said primer and DNA polymerase, thereby the double stranded DNA (X) is prepared. Then, said-double stranded DNA (X) are replicated by repeating said thermal denaturation (I), annealing (II) of the primer and synthesizing (III) of the complementary chain. -17-
