ABSTRACT OF THE DISCLOSURE A method and reagent for reducing errors in any assay systems wherein nicotinamide adenine dinucleotide (NAD) is reduced, the reduced form of NAD being a quanti- tative measure of the sample constituent being measured, and being subject to reaction with any pyruvate and lactic dehydrogenase (LDH) which may be present. Specifically, the method comprises conducting the assay in the presence of oxalic or oxamic acids or their salts, which form an inactive complex with LDH, thereby eliminating interferences due to the presence of any LDH. Glucose and creatine phosphokinase assays are examples of essays wherein the reduced form of NAD is measured, and consequently, the presence of either pyruvate or lactate in the sample will cause errors in the assay. The method can also be employed in assay systems wherein the reduced form of NAD (NADH) is oxidized to NAD. In such systems, the disappearance of NADH is a measure of the constituent being measured so that any interference can cause errors in the assay. An example of such a system is the measurement of plasma ammonia using glutamic dehydrogenase. - 1 -
