The invention provides a novel lytic reagent composition highly selective in its interactions with the cell membranes of white blood cells and also provides a method of using the reagent S composition in a particle analyzing system to effect a significantly improved white blood cell differential determination. The lytic reagent composition is characterized by an ability to selectively shrink the white blood cells into the increasing size order of lymphocytes, basophils, monocytes, cosinophils and neutrophils, and effect a five component separation of these major subpopulations of white blood cells on the histogram of an automated blood analyzer when used in conjunction with a suitable blood diluent as shown in Figures IA to IC.

L'invention concerne une nouvelle composition d'un réactif lytique hautement sélective du point de vue de son interaction avec les membranes cellulaires des lymphocytes et une méthode d'utilisation de ce réactif dans un système d'analyse de particules avec comme résultat une détermination nettement améliorée des différentes classes de lymphocytes. La composition de réactif lytique est caractérisée par le fait qu'elle permet de rétrécir sélectivement les lymphocytes avec comme ordre croissant de la taille: lymphocytes, granulocytes basophiles, monocytes, polynucléaires éosinophiles et granulocytes neutrophiles. L'invention permet de bien distinguer ces cinq classes majeures de leucocytes lors de l'établissement d'un histogramme avec un analyseur de sang automatique utilisé avec un diluant approprié pour le sang comme illustré aux figures 1A à 1C.
