Methods for converting prethrombin-1 to thrombin are disclosed. An aqueous solution of prethrombin-1 is applied to oscutarin-C immobilized on a solid support so as to provide from 500 mg to 4000 mg of prethrombin-1 per mL of the solid support and a contact time between the prethrombin-1 and the oscutarin-C of from 1.8 to 3.5 minutes. The resulting active thrombin may be captured on an ion exchange chromatography medium or an affinity chromatography medium.

L'invention concerne des procédés pour convertir de la préthrombine 1 en thrombine. Une solution aqueuse contenant de la préthrombine 1 est appliquée à de l'oscutarine C immobilisée sur un support solide, ceci permettant la production de 500 mg à 4000 mg de préthrombine 1 par mL du support solide, le temps de mise contact entre la préthrombine 1 et l'oscutarine C étant compris entre 1,8 et 3,5 minutes. La thrombine active ainsi obtenue peut être capturée sur un milieu de chromatographie d'échange d'ions ou un milieu de chromatographie d'affinité.
