The present invention relates to a process for the catalysis of complex reactions of large molecules, namely enzyme-catalyzed reactions, in which, as a rule, undesired secondary or side reactions occur, by means of enzymes bonded to a polymeric support, in which the undesired secondary or side reactions are avoided to the greatest extent by the choice of a pore- free or almost pore-free support material. In particular, the present invention relates to a process for the enzymatic obtainment of biomolecules, preferably of peptides, proteins, oligosaccharides or polysaccharides, from their biologically inactive precursors by means of enzymes bonded to a polymeric support, in particular to a process for the obtainment of insulins or their analogs from the corresponding precursors by means of enzymes bonded to a polymeric support, which, as a result of the choice of a pore-free or almost pore-free support, leads, with avoidance to the greatest extent of secondary or side reactions, to a selective formation of the biomolecules, in particular of the insulins or of the insulin analogs and of the associated valuable substances which can be cleaved to give insulins or their analogs.

L'invention concerne un procédé de catalyse de réactions complexes de grandes molécules, notamment de réactions catalysées par enzymes, où des réactions consécutives ou secondaires indésirables interviennent, à l'aide d'enzymes liées à un support polymère. Selon ce procédé, les réactions consécutives ou secondaires indésirables sont évitées dans une large mesure, grâce à la sélection d'un matériau support sans porosité ou presque. L'invention concerne en particulier un procédé permettant d'obtenir par voie enzymatique des biomolécules, de préférence des peptides, des protéines, des oligosaccharides ou des polysaccharides, à partir de leurs précurseurs biologiquement non actifs, à l'aide d'enzymes liées à un support polymère. L'invention concerne notamment un procédé de préparation d'insulines ou de leurs analogues à partir des précurseurs correspondants, à l'aide d'enzymes liées à un support polymère. Ce procédé permet d'obtenir, grâce au choix d'un support sans porosité ou presque, ce qui évite dans une large mesure la formation de réactions consécutives ou de réactions secondaires, la formation sélective de biomolécules, notamment d'insuline ou d'analogues d'insuline et de substances apparentées, aptes au clivage pour former des insulines ou leurs analogues.
