The invention concerns a method for inducing somatic mutations in vitro or ex vivo, comprising (1): adding to the cells to be mutated, in culture conditions and in a medium suited to said cells, of at least a combination of anti-IgM and anti-CD19 and/or anti-DC21 antibodies, while (2) said anti-IgM antibodies are biotinylated and are subjected to specific aggregation by streptavidin coupled with a support. Advantageously, the mutations are controlled and/or adjusted by inhibiting and reactivating and/or stimulating DNA polymerase pol iota. The invention is in particular applicable to rapid and controlled induction of mutations on the BL2 Burkitt's lymphoma.
L'invention concerne un procédé pour l'induction de mutations somatiques in vitro ou ex vivo, comportant (1) l'adjonction aux cellules à faire muter, dans des conditions de culture et dans un milieu appropriés aux dites cellules, d'au moins une combinaison des anticorps anti-IgM et anti-CD19 et/ou anti-CD21, tandis que (2) lesdits anticorps anti-IgM sont biotinylés et sont soumis à une agrégation spécifique par la streptavidine couplée à un support. Avantageusement on maîtrise et/ou on ajuste les mutations en inhibant et réactivant, et/ou en stimulant l'AND polymérase pol iota. Application notamment à l'induction rapide et maîtrisée des mutations sur le lymphome de Burkitt BL12.
