- 1 - Abstract: The invention relates to an improved method for culturing a yeast transformed by a recombinant plasmid, which is prepared by inserting a hepatitis B virus gene into a plasmid carrying a repressible acid phosphatase promoter of a yeast, by controlling of the promoter and thereby producing the HBs antigen. The method comprises culturing the transformed yeast in a semi-synthetic medium containing yeast extract, an inorganic ammonium salt, an inorganic magnesium salt and either one or both of glucose and saccharose.
