The invention relates to a method for selecting compounds having a modulating effect on the activation state of a dimer of VFT domain proteins expressed in cellular membranes in a measuring medium, said dimer including a first protein and a second protein which are identical or different, wherein the method includes the following steps: (a) marking the first and second proteins in the N-terminal portion of the VFT domain by members of a FRET partner pair, the Förster radius (R0) of said pair ranging between 20 and 55 Å; (b) measuring the FRET signal in the absence and in the presence of the compound to be tested within a predetermined time window; (c) selecting the compound to be tested as a modulating compound if a difference in the FRET signal in the absence and in the presence of the compound to be tested is measured during step (b). The invention can be used in the research for new drugs and new taste modulators.
L'invention a pour objet un procédé de sélection de composés ayant un effet modulateur sur l'état d'activation d'un dimère de protéines à domaine VFT exprimées dans des membranes cellulaires présentes dans un milieu de mesure, ledit dimère étant constitué d'une première protéine et d'une seconde protéine, lesdites protéines étant identiques ou différentes, ce procédé comprenant les étapes suivantes : (a) marquage de la première et de la seconde protéine dans la partie N-terminale de leurs domaines VFT par les membres d'un couple de partenaires de FRET, le rayon de Förster (R0) dudit couple étant compris entre 20 et 55 Å; (b) mesure du signal de FRET en l'absence et en présence du composé à tester dans une fenêtre temporelle déterminée; (c) sélection du composé à tester comme composé modulateur si une différence de signal de FRET en l'absence et en présence de composé à tester est mesurée à l'étape (b). Application : recherche de nouveaux médicaments et de nouveaux modulateurs du goût.
