A method of detecting a nucleic acid which comprises detecting a nucleic acid aptamer formed by hybridizing the target base sequence with a probe by using the binding activity to a ligand as an indication. By using as the nucleic acid aptamer a structure the binding activity of which widely varies depending on a mismatch of a single base (for example, three-way junction), SNP can be detected.
