ABSTRACT A method for detecting and quantitating: any organism containing ribosomal RNA; any member of a large organism category, such as the group of all bacteria; any member of a small organism category such as members of a taxonomic species of bacteria; any member of an intermediate sized organism category; and previously unknown organisms. The method comprises contacting the nucleic acid of the organism whose presence, identification, and quantitation are to be determined, with a marked probe comprising nucleic acid molecules complementary to R-RNA sequences of the said organism, under nucleic acid hybridization conditions, and then determining the degree of hybridization that has occurred. Illustratively, Mollicutes and other bacterial organisms which may be present in a cell-containing medium are detected identified and quantitated by contacting a probe which consists of marked single strand nucleic acid molecules which are complementary to the R-RNA of a member of the class Mollicutes, under hybridizing conditons, with nucleic acid isolated from the cells in the said medium, whereby the probe hybridizes only with R-RNA from any member of the Mollicutes or other bacterial organisms present in the said medium, and then determing the number of Mollicutes organisms present in the medium. Also, illustratively, Trypanosoma organisms (a type of protozoan) which may be present in a mammalian cell containing medium are detected by contacting a probe which consists of a selected marked single strand nucleic acid which is complementary to the R-RNA of a member of the taxonomic group Trypanosoma under hybridization conditions, with nucleic acid isolated from the cells in the said medium, whereby the probe hybridizes only with R-RNA from Trypanosoma present in the said medium, and then determining the number of Trypanosoma organisms present in the medium.
