The invention is located in the field of coagulation diagnosis and relates to an in vitro method for determining von Willebrand factor (VWF) activity in a sample. The method includes the use of a mutant gain-of-function variant of the GPIb.alpha. protein, thus making it possible to dispense with the use of ristocetin, botrocetin or another ristocetin- or botrocetin-equivalent substance. The invention further relates to a method for determining von Willebrand factor (VWF)-cleaving activity of the ADAMTS-13 protease.
L'invention s'appuie sur le domaine du diagnostic de coagulation et concerne un procédé de détermination in vitro de l'activité du facteur von Willebrand (VWF) dans un échantillon. Le procédé comprend l'utilisation d'un variant mutant avec gain de fonction de la protéine GPIba, qui permet de ne pas utiliser ni ristocétine, ni botrocétine ni toute autre substance équivalente à la ristocétine ou à la botrocétine. L'invention concerne en outre un procédé de détermination de l'activité de la protéase ADAMTS-13, clivant le facteur von Willebrand (VWF).
