A method of diagnosing metabolic bone diseases, especially osteoporosis and arthrosis characterized by determining the concentration of osteoclastgenesis inhibitory factor (OCIF) in humor. Monoclonal antibodies recognizing equally both of monomer type and diner type of OCIF. Monoclonal antibodies recognizing selectively diner type of OCIF. And to provide an assay kit for determination of OCIF concentration comprising the aforementioned two antibodies recognizing different epitope of OCIF and having high affinity showing dissociation constant of less than 2 x 10-7 M with antigen. It is useful for a method of diagnosing metabolic bone diseases, especially osteoporosis and arthrosis or for an assay reagent for research thereof.
Cette invention concerne un procédé permettant de diagnostiquer un dysfonctionnement du métabolisme osseux, notamment l'ostéoporose et les maladies des jointures, lequel procédé consiste à mesurer la concentration de facteurs inhibant l'ostéoclastogenèse (FIOC) dans le fluide corporel. Cette invention concerne également un anticorps monoclonal reconnaissant de la même manière les FIOC monomères et dimères, ainsi qu'un anticorps monoclonal reconnaissant sélectivement et uniquement le FIOC dimère. Cette invention concerne en outre des nécessaires d'analyse de FIOC qui contiennent les anticorps monoclonaux des deux types susmentionnés, lesquels reconnaissent les différents épitopes des FIOC et possèdent une grande affinité ainsi qu'une constante de dissociation envers un antigène de 2 x 10<-7> ou moins. Les anticorps et les nécessaires susmentionnés sont utiles afin de diagnostiquer un dysfonctionnement du métabolisme osseux, notamment l'ostéoporose et les maladies des jointures, et peuvent être utilisés dans des réactifs d'analyse en laboratoire, etc.
