A method for specifically diagnosing spinocerebellar ataxia type 2 (SCA2). The method comprises effecting PCR by employing the DNA to be tested as a template and using a first primer comprising a nucleic acid which is hybridizable with a part of the base sequence represented by SEQ ID NO.1 in the Sequence Listing and a second primer comprising a nucleic acid which is hybridizable with a part of the base sequence represented by SEQ ID NO.2 in the Sequence Listing and determining the number of CAG repeat units in the PCR product thus amplified. In this method, the gene of a patient with SCA2 has the number of CAG repeat units of 35 or above while the gene of a normal subject has the number of from 15 to 24, which enables the diagnosis of SCA2.
Ce procédé, destiné à diagnostiquer de manière spécifique l'ataxie spino-cérébelleuse de type 2, comprend les étapes consistant à effectuer une PCR à l'aide de l'ADN à essayer en tant que matrice, à utiliser une première amorce comprenant un acide nucléique qui peut s'hybrider avec une partie de la séquence de base représentée par SEQ ID NO 1 de la liste de séquences, ainsi qu'une seconde amorce comprenant un acide nucléique qui peut s'hybrider avec une partie de la séquence de base représentée par SEQ ID NO 2 de la liste de séquences, puis à déterminer le nombre d'unités de répétition CAG dans le produit PCR ainsi amplifié. A l'aide de ce procédé, on a trouvé que le gène d'un patient atteint d'ataxie spino-cérébelleuse de type 2 présentait un nombre d'unités de répétition CAG de l'ordre de 35 ou plus, alors que le gène d'un sujet normal en présente un nombre compris entre 15 et 24, ce qui permet ainsi de diagnostiquer l'ataxie spino-cérébelleuse de type 2.
