The invention relates to a method and assembly for electrochemically identifying target nucleotide sequences. The method comprises the following steps consisting of: supplying a biological sample that may contain a predetermined target nucleotide sequence; supplying activatable amplification means comprising free nucleotides in order to form replicated target nucleotide sequences; supplying an oxido-reducible compound capable of being inserted during replication between the nucleotides forming the replicated target sequences; and activating the activatable amplification means before applying an electric field to the sample in order to activate the oxido-reducible compound. Said replicated target sequences then cause the inhibition of the electrochemical activity of the inserted oxido-reducible compound, and the presence of the predetermined target nucleotide sequence is determined if the electric current decreases.
L'invention concerne une méthode et un ensemble d'identification électrochimique de séquences cibles de nucléotides. Selon la méthode, on fournit d'abord un échantillon biologique susceptible de contenir une séquence cible de nucléotides déterminée. Puis on fournit des moyens d'amplification activables comportant des nucléotides libres, pour former des séquences cibles de nucléotides répliquées. On fournit également un composé oxydo-réductible apte à venir s'intercaler durant la réplication, entre les nucléotides formant lesdites séquences cibles répliquées. Et on active lesdits moyens d'amplification activables avant d'appliquer un champ électrique audit échantillon pour activer ledit composé oxydo-réductible. Lesdites séquences cibles répliquées provoquent alors l'inhibition de l'activité électrochimique dudit composé oxydo-réductible intercalé et on détermine la présence de ladite séquence cible de nucléotides déterminée si le courant électrique diminue.
