A method for predicting the elution time of a peptide in chromatographic and electrophoretic separations by first providing a data set of known elution times of known peptides, then creating a plurality of vectors, each vector having a plurality of dimensions, and each dimension representing positional information about at least a portion of the amino acids present in the known peptides. A hypothetical vector is then created by assigning dimensional values for at least one hypothetical peptide, and a predicted elution time for the hypothetical vector is created by performing at least one multivariate regression fitting the hypothetical peptide to the plurality of vectors. Preferably, the multivariate regression is accomplished by the use of an artificial neural network and the elution times are first normalized using linear regression.
L'invention concerne un procédé pour prévoir le temps d'élution d'un peptide dans des séparations chromatographiques et électrophorétiques, comprenant la première mise en place d'un ensemble de données de temps d'élution connus de peptides connus, puis la création d'une pluralité de vecteurs, chaque vecteur présentant une pluralité de dimensions, et chaque dimension représentant des informations de position autour d'au moins une partie des aminoacides présents dans les peptides connus. Un vecteur hypothétique est ensuite créé par attribution de valeurs dimensionnelles pour au moins un peptide hypothétique, un temps d'élution prévu pour le vecteur hypothétique est ensuite créé grâce à la réalisation d'une régression multidimensionnelle raccordant le peptide hypothétique à la pluralité de vecteurs. De préférence, la régression multidimensionnelle est effectuée par l'utilisation d'un réseau neural artificiel et les temps d'élution sont normalisés au moyen d'une régression linéaire.
