The invention relates to methods and systems for predicting or estimating the melting temperature of duplex nucleic acids, in the presence of divalent cations, particularly duplexes of oligonucleotides which may be used as, for example, but not limited to primers or probes in PCR and/or hybridization assays. The methods and algorithms use novel formulas, having terms and coefficients that are functions of the particular nucleotide sequence, to estimate the effect of divalent cation salt conditions on the melting temperature.
L'invention concerne des procédés et des systèmes destinés à prédire ou estimer la température de fusion de duplex d'acides nucléiques, en présence de cations bivalents, notamment de duplex d'oligonucléotides qui peuvent être utilisés, par exemple, mais sans y être limité, en tant qu'amorces ou sondes dans des analyses PCR et/ou d'hybridation. Les procédés et les algorithmes utilisent de nouvelles formules, qui comportent des termes et des coefficients qui sont des fonctions de la séquence de nucléotides particulière, pour estimer l'effet des conditions salines des cations bivalents sur la température de fusion.
