This invention relates to a method for evaluating the susceptibility of pharmaceutical drugs to metabolism by a specific cytochrome P-450 isozyme, whichcomprises contacting the sample compound with a reagent composition prepared by adding the specific cytochrome P-450 isozyme to liver microsomes lacking said specific cytochrome P-450 isozyme in a carrier material. More particularly, thisinvention relates to a useful in vitro quantitative assay for drug metabolism by P-450 isozymes such as CYP2D6, CYP2C19, and CYP2A6. This invention also relates to a reagent composition which is useful for said in vitro assay.

L'invention concerne une méthode pour évaluer la sensibilité de médicaments pharmaceutiques à la métabolisation par une isoenzyme spécifique du cytochrome P-450. La méthode consiste à mettre en contact le composé échantillon avec une composition de réactifs préparée par addition, dans un excipient, de l'isoenzyme spécifique du cytochrome P-450 dans des microsomes hépatiques dépourvus de cette isoenzyme. Plus particulièrement, l'invention concerne un essai quantitatif in vitro pour l'évaluation de la métabolisation des médicaments par les isoenzymes du cytochrome P-450 comme CYP2D6, CYP2C19 et CYP2A6. Cette invention a aussi trait à une composition de réactifs qui est utile pour un tel essai in vitro.
