The present invention relates to a process for deriving dendritic cells from mononuclear cells in culture comprising the step of putting in contact type I IFN with said mononuclear cells. Dendritic cells suitable as cellular adjuvants in prophylactic as well as therapeutic vaccination of animal and human beings, are obtainable thereby, after a single step treatment in a brief period of time. Dendritic cells obtainable thereby, pharmaceutical compositions including them, in particular a vaccine comprising said cells as active principle, and a method of treatment of a pathology associated with the presence of an antigen in human beings, are further objects of the invention, as well as a kit for deriving said dendritic cells and a method for the ex vivo expansion of T cells using them.
La présente invention concerne un procédé pour produire des cellules dendritiques à partir de cellules mononucléaires en culture. Ce procédé consiste à mettre en contact des IFN de type I avec lesdites cellules mononucléaires. On peut ainsi obtenir des cellules dendritiques qui servent d'adjuvants cellulaires dans la vaccination prophylactique ou thérapeutique d'animaux ou d'êtres humains, après un traitement en une étape, en une brève période de temps. La présente invention concerne également les cellules dendritiques pouvant être ainsi obtenues, des compositions pharmaceutiques les renfermant, notamment un vaccin dans lequel lesdites cellules constituent le principe actif, un procédé pour traiter une pathologie associée à la présence d'un antigène chez les êtres humains, une trousse permettant de produire ces cellules dendritiques, ainsi qu'un procédé d'expansion ex vivo des lymphocytes T mettant en oeuvre lesdites cellules dendritiques.
