The present invention relates to a method for identifying allergenic proteins and peptides. More specifically, the present invention provides a method for identifying allergenic milk proteins and/or peptides comprising the steps of: providing at least one expression library comprising DNA or cDNA derived from the mammary gland tissue of a lactating mammal, preferably a lactating cow, expressing at least one protein or peptide encoded by said expression library, determining the binding capacity of said at least one protein or peptide to IgE of at least one serum of an individual who is sensitive to mammalian milk,preferably cow's milk, contacting the at least one protein or peptide exhibiting an IgE binding capacity as determined in step c) with basophil cells, eosinophil cells or mast cells and identifying the at least one protein or peptide as being allergenic when said basophil cells, eosinophil cells or mast cells release upon contact with at least one protein or peptide of step d) at least one mediator.
La présente invention porte sur un procédé pour identifier des protéines et peptides allergéniques. Plus spécifiquement, la présente invention porte sur un procédé pour identifier des protéines et/ou peptides du lait allergéniques comprenant les étapes consistant à : fournir au moins une bibliothèque d'expression comprenant de l'ADN ou de l'ADNc issu du tissu de glande mammaire d'un mammifère en lactation, de préférence une vache en lactation, exprimer au moins une protéine ou un peptide codé par ladite bibliothèque d'expression, déterminer la capacité de liaison de ladite au moins une protéine ou ledit au moins un peptide à IgE d'au moins un sérum d'un individu qui est sensible au lait de mammifère, de préférence au lait de vache, mettre en contact la au moins une protéine ou le au moins un peptide présentant une capacité de liaison à IgE comme déterminé à l'étape c) avec des cellules basophiles, des cellules éosinophiles ou des mastocytes et identifier la au moins une protéine ou le au moins un peptide comme étant allergénique lorsque lesdites cellules basophiles, cellules éosinophiles ou mastocytes libèrent lors d'un contact avec au moins une protéine ou au moins un peptide de l'étape d) au moins un médiateur.
