Described are three basic components: (1) methods for the display of functional MHC molecules with covalently attached antigenic peptides on the surface of baculovirus and baculovirus infected insect cells; (2) methods for the identification and physical isolation of baculovirus or baculovirus infected insect cells bearing a displayed MHC/peptide combination that is recognized by a particular T cell antigen receptor; and (3) methods for producing libraries of baculovirus or baculovirus infected insect cells displaying a particular MHC molecule and many different potential antigenic peptides.
Cette invention a trait à trois composantes de base : (1) des méthodes d'affichage des molécules CMH fonctionnelles avec des peptides antigéniques attachés de manière covalente sur la surface d'un baculovirus et de cellules d'insectes infectées par baculovirus, (2) des méthodes d'identification et d'isolation physique de baculovirus ou de cellules d'insectes infectées par baculovirus qui présentent une combinaison de peptides/CMH reconnue par un récepteur antigénique spécifique de lymphocytes T, et (3) des méthodes de production de bibliothèques de baculovirus ou de cellules d'insectes infectées par baculovirus possédant une molécule CMH spécifique et plusieurs peptides antigéniques puissants différents.
