Selective growth inhibitors are identified by (i) providing a semi-solid nutrient medium (e.g., an agar plate) co-seeded with a strain of cells (e.g., bacteria or fungi) that is sensitive to growth inhibitors having a particular mode of action and with a strain that is essentially the same as the sensitive strain except that it is resistant to said growth inhibitors, wherein the sensitive strain is co-seeded in the nutrient medium in an amount substantially in excess of that of the resistant strain; (ii) treating the medium with a test substance; (iii) incubating the treated medium under conditions suitable for the growth of both the sensitive and the resistant strains; and (iv) examining the incubated treated medium for growth inhibition. The observation in (iv) of a zone of no growth except for isolated colonies of the resistant strain is an indication that the test substance is a selective growth inhibitor having the mode of action of interest.
La présente invention a trait à un procédé d'identification d'inhibiteurs sélectifs de croissance comprenant (i) la mise à disposition d'un milieu nutritif semi-solide (par exemple, une boîte de Pétri avec un ensemencement combiné d'une souche de cellules (par exemple, des bactéries ou des champignons) sensible à des inhibiteurs de croissance présentant un mode particulier d'action et avec une souche qui est sensiblement la même que la souche sensible sauf qu'elle est résistante aux dits inhibiteurs de croissance, la souche sensible étant semé conjointement dans le milieu nutritif en une quantité sensiblement supérieure à celle de la souche résistante ; (ii) le traitement du milieu avec une substance d'essai ; (iii) l'incubation du milieu traité dans des conditions appropriées pour la croissance des souches sensible et résistante ; et (iv) l'inspection du milieu d'incubation traité pour l'inhibition de croissance. L'observation à l'étape (iv) d'une zone de non croissance sauf pour des colonies isolées de la souche résistante est une indication que la substance d'essai est un inhibiteur sélectif de croissance présentant un mode d'action d'intérêt.
