It is intended to provide a method whereby a humoral immune response can be induced at an elevated efficiency in constructing an antibody against an antigen protein by gene immunization. By administering a fused gene composed of a gene encoding an antigen protein as a whole or a part thereof attached to another gene encoding a chaperonin subunit or a chaperonin subunit ligation, the fused gene is expressed in the animal and thus a humoral immune response to the antigen protein is induced. As an example of the chaperonin, Escherichia coli GroEL is cited. It is also intended to provide a composition for immunization, a method of producing an antibody, a method of producing a hybridoma and a method of producing a monoclonal antibody.
La présente invention a pour objet une méthode par laquelle une réponse immunitaire humorale peut être induite par production efficace d~un anticorps contre une protéine antigène par immunisation génétique. En administrant un gène mixte composé de tout ou partie d~un gène codant pour une protéine antigène lié à un autre gène codant pour une sous-unité de la chaperonine ou une ligature entre sous-unités de la chaperonine, il est possible de faire exprimer ledit gène mixte par l'organisme de l'animal et ainsi, d'induire une réponse immunitaire humorale à la protéine antigène. La GroEL d~Escherichia coli est citée au titre d~exemple de chaperonine. La présente invention a également pour objet une préparation pour immunisation, une méthode de production d'un anticorps, une méthode de production d'un hybridome et une méthode de production d'un anticorps monoclonal.
