A method for regulating in vivo calcium transport in cardiac muscle of animals suffering from congestive heart failure is disclosed. According to the method, calcium ATPase activity (which decreases as congestive heart failure develops) and cardiac muscle contractility augmented by delivering a gene which operatively encodes the enzyme into the heart. Delivery systems, including but not limited to using adeno-associated viral vectors are provided. Methods for monitoring the expression and effect of the gene product on cardiac performance are also provided.

L'invention concerne un procédé de régulation in vivo du transport de calcium dans le muscle cardiaque des animaux souffrant d'insuffisance cardiaque congestive. Selon ce procédé, l'activité ATPase de calcium (qui décroit lors du développement d'une insuffisance cardiaque congestive) et la constractilité du muscle cardiaque sont augmentées par la délivrance d'un gène qui est opérationnellement codant pour l'enzyme dans le coeur. L'invention concerne des systèmes de délivrance comportant, de manière non exclusive, l'utilisation de vecteurs viraux adéno-associés. L'invention concerne enfin des procédés de surveillance de l'expression et l'effet du produit génique sur les performances cardiaques.
