A method of inducing differentiation from a bone marrow stromal cell to a skeletal muscle cell effectively, simply and reproducibly, comprising the steps of :(a) adding one or more substances selected from the group consisting of a cyclic AMP elevation functioning agent, a cyclic AMP analogue and a cell differentiation survival functioning factor to a culture of a bone marrow stromal cell (note that the bone marrow stromal cell is not treated with a demethylating agent) and performing cultivation; (b) obtaining a myoblast culture by introducing Notch gene and/or a Notch signaling related gene into the cell obtained in the step (a) and performing cultivation (note that the cultivation does not include co-cultivation of a cell into which a gene has been introduced and a cell into which a gene has not been introduced); and (c) adding a Notch ligand to the myoblast culture obtained in the step (b) and performing cultivation.

La présente invention concerne un procédé d'induction de la différenciation d'une cellule stromale de moelle osseuse en une cellule de muscle squelettique de manière efficace, simple et reproductible, comprenant les étapes consistant à :(a) ajouter une ou plusieurs substances sélectionnées parmi le groupe consistant en un agent d'élévation de l'AMP cyclique, un analogue de l'AMP cyclique et un facteur de survie à la différenciation cellulaire dans une culture d'une cellule stromale de moelle osseuse (noter que la cellule stromale de moelle osseuse n'est pas traitée avec un agent de déméthylation) et effectuer une culture ; (b) obtenir une culture de myoblastes en introduisant le gène Notch et/ou un gène associé à la signalisation Notch dans la cellule obtenue à l'étape (a) et effectuer une culture (noter que la culture n'inclut pas la co-culture d'une cellule dans laquelle un gène a été introduit et une cellule dans laquelle un gène n'a pas été introduit) ; et (c) ajouter un ligand Notch dans la culture de myoblastes obtenue à l'étape (b) et effectuer une culture.
