This invention provides a method for inhibiting thrombosis in a patient whose blood is subjected to extracorporeal blood circulation which comprises contacting the extracorporeal circulating blood with a Factor IXa compound in an amount effective to inhibit thrombosis in the patient. The Factor IXa compound may include an active site-blocked Factor IXa compound or Glu-Gly-Arg chloromethyl ketone-inactivated human factor IXa compound. This invention also provides that the effective amount may be from about 0.1 µg/ml plasma to about 250 µg/ml plasma or from about 0.5 µg/ml plasma to about 25 µg/ml plasma. The patient may be subjected to extracorporeal blood circulation during transplant surgery or cardiopulmonary bypass surgery. This invention further provides for a pharmaceutical composition which includes an effective amount of a Factor IXa compound and a pharmaceutically acceptable carrier.
La présente invention a pour objet un procédé destiné à empêcher la thrombose chez un patient dont le sang est soumis à une circulation extracorporelle, ledit procédé consistant à mettre en contact le sang en circulation extracorporelle avec un composé de Facteur IXa, présent en une quantité suffisante pour empêcher la thrombose chez le patient. Le composé de facteur IXa peut comprendre soit un composé de facteur IXa à site actif bloqué, soit un composé de facteur IXa humain inactivé de cétone méthylchoré Glu-Gly-Arg. De plus, selon la présente invention, la quantité suffisante susmentionnée peut être un plasma dont la teneur est comprise entre environ 0,1 µg/ml et environ 250µg/ml ou un plasma dont la teneur est comprise entre environ 0,5 µ/ml et environ 25 µg/ml. Le patient peut être soumis à une recirculation sanguine extracorporelle lors d'une greffe ou d'une opération de pontage cardio-pulmonaire. La présente invention a également pour objet une composition pharmaceutique comprenant une quantité efficace d'un composé de facteur IXa et d'un excipient acceptable sur le plan pharmacologique.
